[Errno 2] No such file or directory when trying to use rc.local or cronjob to run on start up.

I have spi turned on and set up in the config, if I run the python script the code works, however whenever I try to set up a 'run on start up' I get the error on the line: "epd.init(epd.FULL_UPDATE)" I can print the object and it is there but this line it always erroring with [Errno 2] No such file or directory. Any help would be appreciated!
