Here's a tentative plan for the next manuscript:

• Figure 1: Receptor variation drives response heterogeneity
  • Brian's M&M plots showing dose-response profile for differing levels of receptor
  • Single cell PCA plots at a few doses
• Figure 2: Quantifying receptor variability
  • Mean, var, skew of receptors in each cell population
  • The error of each of these estimates will need to be calculated using the replicates
• Figure 3: Quantifying response
  • Mean, var, skew of each population pSTAT5
  • Maybe the wt IL-2/15 in the main figure, with muteins in the supplement
• Figure 4: Moment propagation
  • A cartoon of the approach
  • Fitting to the data and fit parameters
  • Data/fit overlay
• Figure 5: Showing that fitting the higher moments improves the fit
  • Infers the covariance of the receptors?
  • Overall better model fit?
• Figure 6: Exploration/validation
  • Left open for now.
